چکیده

T h : antagonism between tubocurarine and alcuronium chloride on the one hand and neostigmine, physostigmine, edrophonium and ambenonium on the other have been investigated. A quantitative measure of antagonism was obtained on the rat diaphragm preparation and it was found that in general the antagonists were more effective against alcuronium chloride than against tubocurarine. The antagonist drugs potentiated the action of acetylcholine on frog rectus muscle. The muscarinic action of ambenonium was less apparent than that of the other drugs but it caused severe reduction in amplitude of cardiac contraction and arrest in 50 per cent of the preparations. It is concluded that ambenonium is unlikely to prove a satisfactory drug for further investigation in man.

EFFECTS OF CHRONIC ADMINISTRATION OF TWO NEWLY SYNTHESIZED NONDEPOLARIZING NEUROMUSCULAR BLOCKING AGENTS

The potential changes In neuromuscular blockade after chronic (>24 h) administration of sub-paralytic doses of two newly introduced aminosteroidal muscle relaxants were investigated. Org-9426 (rocuronium) and Org-9487 were studied in the in vivo guinea pig gastrocnemius muscle-sciatic nerve preparation. The problems seen after prolonged administration of muscle relaxants are thought to ari...

Effects of cholinergic and adrenergic agents and their antagonists at the neuromuscular junction of the cat extraocular muscles.

D,uke-Elder and Duke-Elder first re ported on the action of choline and nicotine on the mammalian extraocular muscle; this was followed by a study by Brown and Harvey on the cat extrinsic eye muscle. Since then very little investigation has been reported on mammalian extraocular muscle, except by Breinin. Recently, more detailed investigation of cat extraocular muscle has been reported followin...

Effects of neuromuscular blocking agents on central respiratory chemosensitivity in newborn rats.

Neuromuscular blocking agents suppress central respiratory activity through their inhibitory effects on preinspiratory neurons and the synaptic drive from preinspiratory neurons to inspiratory neurons. Central CO2-chemosensitive areas, which partly consist of CO2-excited neurons, in the rostral ventrolateral medulla are thought to provide tonic drive to the central respiratory network and invol...

Clinical pharmacology of neuromuscular blocking agents.

The clinical pharmacology of neuromuscular blocking agents is described. During neuromuscular blockade, succinylcholine attaches to receptors in the motor end plate and depolarizes the neuromuscular junction, making the end plate refractory to acetylcholine. The nondepolarizing relaxants have a structure similar to that of succinylcholine and bind to the same receptors. Instead of depolarizing ...
